The course covers introduction to basic methods used for studying different members of fungi, lichens and bryophytes. The course includes work in laboratory (principles of aseptical work, isolation techniques, cultivation techniques), herbaria and libraries, preparation of culture media and macro- and microphotographs, data sampling, storing and evaluating, dying methods, material documentation, etc. Search for relevant literature and preparation of manuscript, theses, posters, etc., is also included.

The course runs in English if at least one non-Czech student is enrolled.

Common techniques:

Preparation of natural media as well as agar media. Sterilization and disinfection. Media for cultivation and media for determination.

Special techniques in mycology:

Isolation methods of cultivated fungi from different substrata. Methods of cultivation. Preservation and storage of saprotrophic fungi in collection. Preparation for optical microscopy: monosporic isolation, agar blocks, microscope slides with agar, moist chambers, staining, preparation of permanent slides etc.

Ascomycetes - direct study of natural material.

Methods for study of plant pathogens: collection, preservation, determination, inoculation experiments. Methods of pathometry. Analyses of important fungal pathogens populations.

Special techniques in bryology.

Collecting of bryophytes and preparation of herbarium material. Determination of bryophytes. Review of basic bryological literature, exsiccates. Cultivation methods. Cytology - study of mitotic and meiotic karyotypes.

Special techniques in lichenology.

Preparation of herbarium material. Chemistry of lichens (spot reactions, extraction and recrystalisation of lichen substances).

Technique for microscopy.

Microscope calibration, phase contrast, DIC.

Documentation techniques.

Drawings, nondigital photomicrography. Black and white photos developing. Photografy of cryptogamologic micro- and macroobjects.

Literature, evaluation of data.

References [rules]. Searching of literature. Preparation of manuscripts. Typography [not only by computer]. Proof-reading. Internet. Library. Basic literature for determination. Evaluation of data by statistic methods. Graphs.
