Prvním z cílů navrhovaného doktorského projektu je příprava a validace fenotypické eseje pro vysoko-kapacitní testování knihovny malých molekul na inhibici dráhy syntézy proteasomu závislé na Nrf1 transkripčním faktoru. Touto esejí bude následně otestována knihovna malých molekul. Pozitivní hity budou validovány na inihibici proteinu DDI2 a dále charakterizovány metodami jako povrchová plasmonová resonance (SPR) či isotermální titrační kalorimetrie (ITC). 3D struktura nejvíce potentních inhibitorů v komplexu s DDI2 proteasou bude případně vyřešena pomocí rentgenové krystalografie. V neposlední řadě budou případné inhibitory DDI2 proteasy použity k hlubšímu popsání funkce DDI2 v buňce.
Preliminary scope of work in English
Initial aim of the proposed doctoral project is the validation of the reporter phenotypic assay for the high-throughput screening of compound libraries for hits affecting Nrf1 regulatory function in the synthesis of new proteasome units.Secondly, binding of validated DDI2 inhibitors will be characterized by the surface plasmon resonance (SPR) technology and isothermal titration calorimetry. A 3D structure of the complex of recombinant DDI2 protease and the most potent ligand/s will be solved and characterized. Last but not least, DDI2 ligands will be used as tools to completely uncover DDI2 function in Nrf1- and Nrf3-dependent pathways.
