Cílem práce (rešerše) je vyhledat a přehledně shrnout publikované údaje o využití permeabilizovaných buněk a hrubých buněčných extraktů k experimentálnímu studiu polymerace aktinu a dynamiky aktinového cytoskeletu vůbec (zejména v kombinaci s pokročilými metodami fluorescenční a konfokální mikroskopie). Výstupem by měl být teroetický návrh postupu použitelného pro rostlinný materiál (klíčící pyl, popřípadě protoplasty pletivových buněk, buněčné kultury) a proveditelného v podmínkách našeho pracoviště. Praktická aplikace získaných poznatků a navržených postupů se může stát součástí navazující diplomové práce.
Preliminary scope of work in English
Aim of this work is to find and systematically summarize published methods using permeabilized cells and crude cell extracts to experimentally study actin polymerization and actin cytoskeleton dynamics in general (esp. in combination with the advanced methods of fluorescent and confocal microscopy). This should result in a theoretical proposal of the method appolicable for the plant ,material (germinating pollen, protoplasts, cell cultures) in the conditions of our Department. Practical application of such knowledge and methods prosposed might become a part of the putative future master thesis work.
