Fluorescenční super-rozlišovací mikroskopie způsobila revoluci ve studiu buňek a tkání. V této práci se změříme na metody a data z TIRF-SIM mikroskopie (Total internal reflection fluorescence - structured illumination microscopy) dosahující prostorového rozlišení 100nm, které je dvakrát větší než rozlišení standardní světelné mikroskopie s širokým zorným polem. Vysoké prostorové a časové rozlišení při nízkém osvětlovacím výkonu řadí TIRF-SIM mikroskopii mezi přední metody ke sledování dějů v živých buňkách. Sledování procesů v živých buňkách však klade vysoké nároky na analýzu pořízených dat a současná biomedicínská komunita postrádá automatické metody zpracování obrazu.
Úkolem řešitele bude vyvinout nové metody zpracování časosběrných dat, které umožní automaticky sledovat dynamické děje v buňkách a statisticky vyhodnocovat jejich chování, a dát tak biomedicínské komunitě základní nástroj na objektivní analýzu experimentů s TIRF-SIM mikroskopem. |

Over the last two decades, numerous fluorescence super-resolution imaging techniques have been invented. This super-resolution revolution has transformed the way cells and tissues are studied. In this work we will focus on methods and data obtained from Total internal reflection fluorescence - structured illumination microscopy (TIRF-SIM). This method offers up to 100 nm spatial resolution, which is twice as much compared to conventional wide-field microscopy. The high spatial and temporal resolution and the low illumination power of TIRF-SIM are optimal for live-cell imaging which is challenging with other super-resolution techniques. However, this type of live-cell imaging data represents a significant challenge for data analysis, and the biomedical community currently lacks objective and automatic methods. Hence to bridge this gap, the student will develop novel image processing approaches for this type of time lapse movies. In summary, this work will provide the biomedical community with an essential tool for unbiased analysis of experiments studied with TIRF-SIM.
