Určení antero-posteriorní polarizace končetiny obratlovců je tradičním tématem vývojové biologie a učebnicovým příkladem role organizátoru okolních tkání. Klasické transplantační experimenty ukázaly, že zadní část končetinového pupene (tzv. zone of polarizing activity, ZPA) řídí polaritu budoucí končetiny. Pokud je ZPA transplantována do přední části končetinového pupene, vytvoří se končetina, jejíž palcová strana se vyvine ve stranu malíkovou. Dojde tedy k zrcadlovému obrazu zadní části končetiny. Kromě končetin existují i další tělní extremity s antero-posteriorní polarizací (například vnější žábry), u nichž by se podobný mechanismus dal předpokládat. Dosud však nebyl prokázán. Tento projekt by se měl věnovat zhodnocení mechanismu určení antero-posteriorní polarizace končetinového pupene a zjistit, zda by daný mechanismus mohl působit i v případě jiných tělních extremit. U zadaného bakalářského projektu se předpokládá následná návaznost v podobě projektu diplomového.

Preliminary scope of work in English

Development of the antero-posterior polarity in the vertebrate limb represents a traditional topic of developmental biology and a text-book example of the role of the organizer of surrounding tissues. Classical transplantation experiments have shown that the posterior part of the limb bud (the so called zone of polarizing activity, ZPA) regulates the polarity of the prospective limb. If the ZPA is transplanted into the anterior part of the limb bud, this portion of the limb, normally fated to become the thumb side, transforms into the little finger side. The limb then displays a little finger mirror image. Besides limbs, there are other body appendages with antero-posterior polarity (e.g. external gills), where similar patterning mechanism would be expected. However, it has not been demonstrated yet. This project aims at evaluation of mechanisms of establishment of antero-posterior polarity of the limb bud and suggesting, whether similar mechanism could play role in case of other body appendages.
