N6-Methyladenosin (m6A) je často se vyskytující modifikací RNA. Výzkum v posledních letech ukazuje, že metylace RNA je citlivě koordinovaným procesem, který může mít řadu významných regulačních funkcí. RNA-demetyláza FTO je důležitým enzymem regulujícím metylaci adenosinu. Cílem této práce je provést knockdown genu pro FTO metodou siRNA silencingu a porovnat jeho vliv na vybrané buněčné funkce u neurálních a astrocytálních buněčných linií s působením nedávno syntetizovaného farmakologického inhibitoru FTO.

N6-Methyladenosine (m6A) is a ubiquitous RNA modification. Recent research suggests that RNA methylation is a tightly regulated process with important regulatory functions. RNA-demethylase FTO is an important enzyme regulating adenosine methylation. The aim of this thesis is to target FTO by siRNA silencing and compare the effects of FTO knockdown to the effects of recently synthetised FTO inhibitor on astrocytic and neuronal cell lines.
