Methylace adenosinu je ubikvitní modifikace RNA, která ovlivňuje „osud“ RNA, čímž může regulovat obrovský počet buněčných procesů. Zatímco methyltransferázy jsou zodpovědné za methylaci adenosinu, demethylázy (jako gen FTO nebo skupina genů ALKBH) se jeví jako klíčové v roli regulátora epigenetické modifikace RNA. Nedávný výzkum naznačuje, že gen FTO je zodpovědný za onemocnění, jakými jsou například obezita, metabolický syndrom, kognitivní porucha či Alzheimerova choroba. Dlouhodobým cílem naší laboratoře je charakterizovat roli či chování genu FTO v mozku a posoudit jeho potenciál jako molekulárního cíle pro léčbu nemocí a poruch metabolismu a Alzheimerovy choroby. Cílem tohoto projektu je posoudit roli FTO v ontogenetickém vývoji myšího mozku v neurogenezi indukované existencí obohaceného prostředí a určit cirkadiální profil mozkové exprese FTO proteinu.

Adenosine methylation is a ubiquitous RNA modification, which affects RNA fate and thus may regulate a vast number of cellular processes. While methyltransferases are responsible for adenosine methylation, demethylases such as FTO or the ALKBH family appear to have a key role in the regulation of this epigenetic RNA modification. Recent research suggests that FTO is implicated in obesity, metabolic syndrome, cognitive impairment and Alzheimer’s disease. The long-term goal of our laboratory is to characterize the role of FTO in the brain and to assess its potential as a molecular target in the treatment of Alzheimer’s disease and metabolic disorders. The objectives of this project are to assess the role of FTO in ontogenetic development of the rat brain, in neurogenesis induced by housing in enriched environment and to determine the circadian profile of FTO protein expression the brain.
