Modelový mech Physcomitrella patens umožňuje využít homolgní rekombinace ke studiu funkce vybraných genů. Tento metodický přístup bude využit ke studiu funkce vybraných podjednotek poutacího komplexu exocyst - zvl. SEC3 - v regulaci buněčné polarity a morfogeneze, ale také v reakci na stress.
Preliminary scope of work in English
Physcomitrella patens is quickly gaining popularity as a model organism in the scientific community. Nowadays it is widely used to elucidate the secrets of evolution of land plant. It has unique property of homologous recombination which makes it excellent organism for reverse genetics approaches. This topic deals with the characterization of exocyst complex (EXO70s, SEC3s and SEC6 subunits) in P.patens. It focuses on making knockout mutants in these subunits and studying the phenotype at morphological and developmental levels, using various molecular biology and biochemical approaches (Sothern and Western Blots, RT-PCR, PCR, cloning technique, light and confocal microscopy etc). Our preliminary data shows interesting observations, with growth arrest at bud level in Ppsec6 and female sterility in case of Ppexp70.3d mutants. We will make similar single and double mutants for SEC3 gene in moss, characterize them and tag them with fluorescent markers. In parallel we will also do some stress experiments to check the response of these mutants, as while colonizing the land, tackling stresses would have proven real challenge for plants.
