Preliminary scope of work
Hlavním cílem práce je analýza části biosyntetické dráhy manumycinových antibiotik zodpovědné za tvorbu polyketidových řetězců. Jedná se především o analýzu genů a proteinů ovlivňujících výběr prekurzorů řetězců a jejich délku. Hlavními metodikami bude příprava mutantních kmenů technikami genové disrupce, exprese heterologních genů v mutantech a analýza produkovaných metabolitů.
Preliminary scope of work in English
The main aim of the work is the analysis of a part of manumycin antibiotics biosynthetic route responsible for the formation of polyketide chains. This involves in particular the analysis of genes and of proteins implicated in the selection of metabolic precursors and in determining of the chain lengths. The main methods will be preparation of mutant strains with the help of gene disruption, expression of heterologous genes in the mutants, and analysis of the metabolites produced by the recombinant strains.
