Součastí diplomové práce bude vyhledání dosavadních poznatků o hemoproteinech, v nichž heme slouží jako signál (hem-detekující proteiny) nebo jako detekční místo (plyn-detekující proteiny). Heterologní expresí bude navíc připraven modelový senzorový protein obsahující hem. Bude ověřeno, zda tento protein obsahuje svou prostetickou skupinu tj. hem a tedy se nachází ve své holo-formě. Dále budou studovány vlastnosti tohoto modelového proteinu.
Preliminary scope of work in English
A model heme-containing sensor protein will be prepared by heterologous expression in bacterial system. It will be verified whether the protein contains its prosthetic group, ie heme and thus is in its holo-form. The biochemical properties of this model protein will be studied.
