Současné výzkumy potvrzují kardioprotektivní vliv na srdce v průběhu chladové aklimace. Cílem diplomové práce bude zjistit, zda roční období (zima vs léto) má vliv na odolnost mitochondrií během chladové aklimace. Jako hlavní metodický přístup bude použit mitochondriální swelling, v průběhu akutní (1, 3 a 10 dnů) a chronické (5 týdnů) aklimace. Z potkaních samců, které budou vystaveny chladu při 8 stupních, bude vyjmuto srdce a z levé komory budou izolovány mitochondrie. Izolované mitochondrie pak budou měřeny metodou swellingu, kdy se postupným zvyšováním koncentrace vápníku pozoruje odolnost mitochondrií.
Preliminary scope of work in English
Recent research confirms cardioprotective effects on the heart during cold acclimation. The aim of the diploma thesis will find out if the season (winter vs summer) has an influence on mitochondrial swelling during cold acclimation. The main methodological approach will be used mitochondrial swelling during acut and chronic aclimation. The heart will be removed from male rats exposed to cold at 8 degrees and mitochondria will be isolated from the left ventricle. Isolated mitochondria will then be measured by the swelling method, where the resistance of mitochondria is observed by gradually increasing the calcium concentration.
