Přenos mikrobioty mezi matkou a novorozencem je komplexní proces zahrnující několikero příspěvků. Jedním z potenciálně možných je přenost prostřednictvím kojení zahrnující transfer prostřednictvím antigenprezentujících buněk. Pro pochopení příspěvku transferu anaerobním bakterií ze střeva do mléčné žlázy a následně rezultující v kolonizaci novorozence bude využit recentně etablovaný model bezmikrobní MHC II. - EGFP knock-in myši. Budou využity fluorescenčně tagované bakterie, metody pokročilé fluorescenční histologie, mnohobarevné průtokové cytometrie i charakterizace mikrobiomu pomoci NGS.
Preliminary scope of work in English
Microbiota transfer between mother and newborn is a complex process involving several contributions. One potentially possible is breastfeeding involving transfer through antigen-presenting cells. Recently established model of germ-free MHC II/EGFP will be used to understand the contribution of anaerobic bacteria transfer from intestine to mammary gland and subsequently resulting in neonatal colonization. Fluorescence-tagged bacteria, methods of advanced fluorescence histology, multicolour flow cytometry and microbiome characterization by NGS will be used.
