| Hlavním cílem této práce bude studium struktury komplexů proteinu 14-3-3, proteinkinasy CaMKK1 a kalmodulinu (Ca2+/CaM). Práce bude zahrnovat: - Exprese a purifikace proteinu 14-3-3, fosforylované CaMKK1 a CaM - Ověření fosforylace CaMKK1 pomocí phos-tag SDS-PAGE a MS - Rekonstituce komplexů (14-3-3/CaMKK1 a 14-3-3/CaMKK1/CaM) in vitro a příprava vzorků pro AUC a SAXS experimenty - Modelování struktury komplexů 14-3-3/CaMKK1 a 14-3-3/CaMKK1/CaM na základě dostupných strukturních a SAXS dat |
