Preliminary scope of work

Práce se bude zabývat jednak metodami izolace exosomů z buněčných kultur, používaných při studiu infekce myšího polyomaviru.
Především se jedná o ultracentrifigaci a izolaci exosmů v hustotním gradientu, následnou vizualizaci exosomů pomocí elektronové
mikroskopie a ověření přítomnosti exosomových proteinových markerů. Dalším cílem bude ověřit hypotézu, že exosomy produkované buňkami,
které jsou infikované polyomavirem, transportují virové proteiny a/nebo nukleové kyseliny do okolních buněk.

Preliminary scope of work in English

The thesis will deal with methods of exosome isolation from cell cultures used for studies of polyomavirus infection. We will use ultracentrifugation and exosome isolation in density gradient, followed by visualization of exosomes by electron microscopy and verification of presence of exosomal protein markers. The next objective is to verify the hypothesis, that exosomes produced by cells infected with polyomavirus transport viral proteins and/or nucleic acids to the surrounding cells.
