Regulace alternativního pre-mRNA sestřihu zahrnuje několik regulačních úrovní. V naší laboratoři studujeme, jak struktura a modifikace chromatinu ovlivňují rozhodnutí o alternativním sestřihu. Před nedávnem jsme ukázali, že promotorové sekvence modulují alternativní sestřih pomocí acetylace histonů. Tento projekt přímo navazuje na naše předchozí výsledky a klade si za cíl otestovat hypotézu, že transkripční enhancery ovlivňují alternativní sestřih. Využijeme CRISPR/Cas9 technologii pro odstranění transkripčních enhancerů lidského genu FN1 a za pomoci RT-qPCR budeme analyzovat sestřih alternativních kazetových exonů tohoto genu.
Preliminary scope of work in English
Regulation of alternative pre-mRNA splicing involves several regulatory levels. Our laboratory has been studying how chromatin structure and modification influences alternative splicing decisions. We have shown that promoter sequences modulate alternative splicing via changes in histone acetylation. This project is direct continuation of this effort. We aim to test the hypothesis that transcription enhancers influence alternative splicing. We will utilize CRISPR/Cas9 approach to delete transcription enhancers of human FN1 gene and analyze splicing pattern of FN1 alternative cassette exons by RT-qPCR.
