Syntetické hydrogely připravené polymerizací hydrofilních monomerů připomínají svými vlastnostmi živé tkáně, jsou často biokompatibilní a poskytují vhodné prostředí pro kultivaci buněk v modelových experimentech i léčbě. Botnání gelů ve vodných prostředích a jejich mechanická pevnost ovlivňují adhezi a chování kultivovaných buněk a určují, zda materiál splní náročné požadavky na aplikovatelnost. Další fyzikální vlastnosti gelů jako jsou jejich optická kvalita (index lomu), mikrostruktura (porozita) jsou dané kompozicí polymerizujícího systému a podmínkami přípravy. Cílem práce je nalézt (kvantitativní) souvislosti mezi strukturou a chováním gelu při botnání a mechanickém statickém i dynamickém namáhání. Práce zahrnuje charakterizaci struktury gelů připravených podle původního postupu v Ústavu makromolekulární chemie a charakterizaci jejich mechanického chování. Cílem práce je nalézt vhodné teoretické přístupy pro popis elastického chování těchto gelů založených na statisticko-mechanických modelech homogenních a poroelastických materiálů a verifikovat je experimentálně na zařízení, které je na KMF MFF popřípadě na ÚMCH AV ČR k dispozici.
Preliminary scope of work in English
Synthetic hydrogels, due to their properties resembling living tissues, provide accurate environment for hosting cell cultures and thus attract interest of researchers working in both, the materials science and biology fields. The important parameters of hydrogels in such applications, besides chemical compatibility, are content of aqueous liquids in swelling equilibrium and swollen material strength and deformability. Other parameters are porosity and optical appearance. These parameters can be controlled by selecting starting components and by topology of the polymer network structure. The work aims at investigation of rules governing the swelling behavior and deformation responses of synthetic hydrogels in the connection with their polymer network structure. The work includes experimental characterization of hydrogels synthesized in the Institute of Macromolecular Chemistry of the Academy of Sciences of Czech Rep. in Prague - mainly swelling and mechanical responses, for instance using rheometry. The method of hydrogels preparation will also include laser cutting technique. The theoretical approaches for description of hydrogel behavior based on physico-chemical models of homogeneous and poroelastic gels will be established and experimentally verified.
