Preliminary scope of work in English
The architecture of root system and functional/anatomical properties of its component roots determine the mode of plant-soil interaction. Root system architecture significantly affects plant’s ability to conquer mineral nutrients or water but also to survive stress conditions such as drought, salinity, presence of phytotoxic compounds within the rhizosphere. Root morphology is determined mainly via branching of the root system, which is carried out by formation of new laterals and their subsequent growth via activity of root apical meristem and elongation of produced cells.
Proposed work is focused on identification of new regulatory elements of lateral root formation using collection of gene-trap lines, which were selected according to expression pattern related to lateral roots and their development (cooperation with Dr. Joseph Dubrovsky, IBT UNAM, MX). Identification of affected genes was/will be carried out with aid of TAIL PCR in lines selected during this forward screening. Several genes were already acquired and their further functional characterization is expected or in progress. The primary task of proposed position is further proceeding identification of new candidate genes and their subsequent functional characterization. Their role in root development will be followed during phenotyping of insertion mutants from publically available collections (SAIL, SALK, GK, etc.) or after overexpression/interference modulation of activity of particular gene. Methods of light microscopy (confocal, fluorescence), quantitative image analysis of root system, and selected methods of molecular biology (cloning, transformation of Arabidopsis, usage of reporter genes in translational or transcriptional fusion, etc.) will be combined during the work. The work will be started with MGT 767 with expression specific for early lateral root primordia development and lines MGT450, MGT 260 and MGT 22 lines with root specific promotor activity.
