Preliminary scope of work

Příprava konstruktů kódujících extracelulární čast leukocytárních receptorů (AICL a NKR-P1C^BALB). Heterologní exprese zmíněných receptorů v E. coli a optimalizace jejich in vitro složení. Ověření správnosti sbalení receptorů pomocí gelové filtrace a hmotnostní spektrometrie. Stanovení disulfidických můstků u získaných bílkovin.

Preliminary scope of work in English

Preparation of constructs coding extracellular part of leucocyte receptors (AICL a NKR-P1C^BALB). Heterologous expression of these receptors in E. coli and optimalization of their refolding. Confirmation of the folding correctness using size-exclusion chromatography, and mass spectrometry. Finally, characterization of disulphide bond formation using mass spectrometry.
