Mitochondriální procesující peptidasa (MPP) nebo její redukované formy vyskytující se v hydrogenosomech a mitosomech patří mezi esenciální enzymy vyskytující se ve všech eukaryotech.
MPP se skládá ze dvou podobných, ale funkčně odlišných podjednotek, alpha-MPP a beta-MPP, jejichž vzájemná spolupráce je pro funkci enzymu nutná. Univerzální funkce tohoto enzymu je v rozeznání a odštěpení velkého množství presekvencí preproteinů, lišící se délkou i primární sekvencí aminokyselin. Kombinací biochemických, molekulárně-genetických a fyzikálních metod byla pozorována změna konformace alpha-MPP po navázání substrátu, a tak byl částečně objasněn mechanismus rozpoznávání široké škály substrátů. Znalost mechanismu fungování MPP poskytuje poznatky i o jejích redukovaných formách, mitosomální procesující peptidase z Giardia (GPP) a hydrogenosomální procesující peptidase (HPP), vyskytujících se převážně v jednobuněčných parazitech. Funkce těchto peptidas, množství substrátů i vlastnosti presekvencí, které rozeznávají, se během evoluce přizpůsobily nižším požadavkům parazitů. |

Mitochondrial processing peptidase (MPP) or its reduced forms occuring in hydrogenosomes and mitosomes are ranked among essential enzymes of all Eucarya.
MPP consists of two similar but functionally different subunits, alpha-MPP and beta-MPP. The cooperation of these subunits is necessary for enzyme function. A universal function of this enzyme is in recognition and cleavage of great number of preprotein presequences, which differ in length and amino acid sequence. The conformational change of alpha-MPP after substrate binding was apparent after using the combination of biochemical, molecular-genetical and physical methods. A mechanism of recognition of wide variety of substrates was therefore partly explained. Knowledge of MPP function mechanism also gives information about its reduced forms, Giardia mitosomal processing peptidase (GPP) and hydrogenosomal processing peptidase (HPP), occuring mainly in unicellular parasites. Functions of these peptidases, quantity of substrates as well as properties of recognized presequences adapted to lower requirements of parasites. |
