Preliminary scope of work
Práce se bude týkat výzkumu nových potencionálních léčiv cílených na mitochondrii lidského parazitického prvoka Leishmania. V rámci práce bude využito různých metod, které budou mít za cíl charakterizovat účinek vybraných látek na patogena, například stanovení inhibiční konstanty léčiva proti parazitovi a lidským buňkám, záznam růstové křivky parazita při podání léčiva, stanovení mitochondriálního membránového potenciálu, množství reaktivních forem kyslíku, buněčného dýchání atd. Kromě experimentální části práce, bude mít student/ka za cíl také shrnout dosavadní poznatky o studovaném parazitovi a současných možnostech léčby.
Preliminary scope of work in English
The work will involve the investigation of new potential drugs targeting the mitochondria of the human parasitic protozoan Leishmania. The work will use various methods to characterize the effect of selected compounds on the pathogen, such as determination of the inhibition constant of the drug against the parasite and human cells, recording of the parasite growth curve upon drug administration, determination of mitochondrial membrane potential, amount of reactive oxygen species, cellular respiration, etc. In addition to the experimental part of the work, the student will also aim to summarize the current knowledge of the studied parasite and current treatment options.
