Fluorescenční mikroskopie je nepostradatelným nástrojem v biofyzice pro zobrazování vzorků buď jejich autofluorescencí, nebo pomocí fluorescenčních značek jako jsou barviva nebo fluorescenční proteiny. Detekce fluorescence nicméně ze své podstaty poskytuje informace pouze o konečném, emisním stavu molekul po fotoexcitaci. Oproti tomu metody ultrarychlé spektroskopie typu transientní absorpce umožňují sledovat počáteční stav molekul po absorpci a následnou excitační dynamiku. Ultrarychlá nelineární spektroskopie však obvykle vyžaduje objemové vzorky a je detekována koherentně. Nedávno jsme vyvinuli nový způsob měření transientní absorpce pomocí detekce fluorescence vzorku (Malý et Brixner, Fluorescence-Detected Pump–Probe Spectroscopy, Angew. Chem. Int. Ed. 60, 18867 (2021)). Tato práce je součástí projektu FluoTRAM, ve kterém implementujeme naši techniku ve fluorescenčním mikroskopu. Náplní práce je generace čerpacího (pump) svazku a jeho modulace na polovině opakovací frekvence, kombinace pump a probe pulzů v mikroskopu a nalezení nelineárního (pump–probe) signálu.

Fluorescence microscopy is an indispensable tool in biophysics for imaging samples either by their autofluorescence or by fluorescent labels such as dyes or fluorescent proteins. However, fluorescence detection inherently provides information only about the final, emission state of molecules after photoexcitation. In contrast, ultrafast spectroscopy methods such as transient absorption allow monitoring of the initial state of the molecules after absorption and the subsequent excitation dynamics. However, ultrafast nonlinear spectroscopy usually requires bulk samples and is detected coherently. Recently, we have developed a new way to measure transient absorption by detecting sample fluorescence (Malý et Brixner, Fluorescence-Detected Pump-Probe Spectroscopy, Angew. Chem. Int. Ed. 60, 18867 (2021)). This thesis is part of the FluoTRAM project, in which we implement our technique in a fluorescence microscope. The scope of this work is the generation of the pump beam and its modulation at half the repetition frequency, combination of pump and probe pulses in the microscope, and isolation of the nonlinear (pump–probe) signal. |
