Preliminary scope of work

Náplní práce bude rekombinantní exprese rozpustné formy receptoru CD69 (tzv. ranný aktivační antigen lidských lymfocytů), jeho dvou mutovaných forem a jeho ligandu galektinu-1, a studium jejich vzájemné interakce s cílem jejího strukturního popisu. Rekombinantní exprese proteinů v bakteriálním systému i v lidské buněčné linii HEK293, purifikace proteinů a charakterizace jejich vzájemné interakce pomocí biofyzikálních metod, krystalizace proteinů a řešení struktury jejich komplexu pomocí proteinové krystalografie.

Preliminary scope of work in English

The aim of the thesis will be the recombinant expression of the soluble form of the CD69 receptor (the so-called early activating antigen of human lymphocytes), its two mutant forms and its ligand galectin-1, and the study of their interaction with the aim of its structural description. Recombinant expression of proteins in the bacterial system and in the human cell line HEK293, purification of proteins and characterization of their interaction by biophysical methods, protein crystallization, and structure determination by protein crystallography.
