Diplomový projekt se zabývá charakterizací organizátoru levo-pravé asymetrie u ne-teleostní paprskoploutvé ryby jesetera malého (Acipenser ruthenus) a (případně) ocasatého obojživelníka axolotla mexického (Ambystoma mexicanum), který se nazývá gastrocélní střešní deska a nachází se na stropě archenteronu gastrulujícího embrya. Projekt si klade za cíl popsat vznik, stavbu, funkci a zánik tohoto orgánu pomocí histologických, imunofluorescenčních, expresních a funkčních přístupů s cílem zhodnotit evoluci organizátorů levo-pravé asymetrie u obratlovců, především pak evoluci Kupfferova váčku u teleostních skupin paprskoploutvých ryb.
Preliminary scope of work in English
The diploma project aims at characterization of the organizer of left-right asymmetry in a non-teleost ray-finned fish, the sterlet sturgeon (Acipenser ruthenus) and (potentially also) a salamander, the Mexican axolotl (Ambystoma mexicanum). This organizer is called "gastrocoel roof plate" and is present on the archenteron roof of the gastrulating embryo. The core of this project lies in describing the rise, structure, function and fall of this organ utilizing histological, immunofluorescent, expression and functional approaches with the aim of evaluating the evolution of the left-right organizer in vertebrates, especially the evolution of Kuppfer's vesicle of teleost ray-finned fishes.
