Nedávno jsme zjistili, že aklimace na mírný chlad vyvolává kardioprotektivní fenotyp u potkana, který je doprovázen zvýšenou odolností mitochondrií k vápníkovému přetížení. Další výsledky naznačují, že kardioprotekce se vyvíjí v časných fázích průběhu aklimace. Proto cílem této diplomové práce je prokázat či vyvrátit, že kardioprotektivní fenotyp vyvolaný aklimací na mírný chlad souvisí s aktivací pro/proti apoptotických drah. Předmětem diplomové práce bude stanovení exprese pro a proti-apoptotických proteinů pomocí western blotu, připadně ELISA kitu, v průběhu vývoje kardioprotektivního režimu chladové aklimace (K, 1- 10 dní, tj. ve 4 skupinách 1D, 3D, 10D), a dále u kontrolní skupiny, po 5 týdnech chladu a po 14 denní regresi.
Preliminary scope of work in English
We recently found that mild cold and a cardioprotective phenotype in the rat, which is accompanied by increased mitochondrial resistance to calcium overload. Other results indicate that cardioprotection only occurs during the early phase of acclimation. Therefore, the aim of this thesis is to prove or disprove that the protective phenotype induced by acclimatization to mild cold is related to the activation of pro/anti-apoptotic pathways. The subject of the thesis will be the determination of the expression of pro- and anti-apoptotic proteins using a western blot, possibly an ELISA kit, during the development of the cardioprotective regimen of cold acclimation (K, 1-10 days, i.e. in 4 occurrences of 1D, 3D, 10D), and further in the control group, after 5 weeks of cold and after 14 days of regression.
