Žlučové kyseliny jsou aktivátorem pro membránový receptor žlučových kyselin (TGR5). Receptor je spřažený s G-proteinem, který je exprimován v epitelu žlučníku, střeva, ale také v kosterním svalstvu a v neposlední řadě také v CNS – např. v enterické nervové soustavě, jež se nachází v trávicím ústrojí obratlovců. Stimulací TGR dochází k produkci glukagon-like peptidu 1, a k ovlivnění metabolismu glukózy. Tento receptor se navíc nachází i v buňkách štítné žlázy a hnědé tukové tkáně, kde žlučové kyseliny vykazují nepochybné metabolické účinky. Cílem navrhované disertační práce je syntéza nových steroidních modulátorů TRG5. |
Preliminary scope of work in English |
The TGR5 receptor was identified as G-coupled protein receptor specific for bile acids. TGR5 gene expression is widely distributed, including gall bladder, intestines, muscles, and the enteric nervous system within the gut wall. The protection against insulin resistance has been attributed to intestinal secretion of glucagon-like peptide 1 induced through the activation of TGR5. Moreover, recent evidence indicate that bile acid administration augmented energy expenditure in mice, and that these effects were mediated via the TGR5. The main aim of the proposed Thesis is synthesis of novel steroidal compounds as modulators of TGR5 and their structute-activity relationship. |
