Preliminary scope of work

Střevní mykobiota je dle posledního výzkumu považována za důležitý faktor ve vývoji některých onemocnění, k nimž patří i nespecifické střevní záněty (ulcerózní kolitida a Crohnova choroba).
Bakalářská práce má shrnout poznatky o mykobiomu trávicího traktu člověka. Práce má podat přehled o diverzitě hub, faktorech určujích složení jejich společenstev (věk, strava, nemoci, pohlaví, orgán), interakce s ostatními mikroby. U hlavních houbových druhů bude shrnuta jejich ekologie a podrobněji interakce s okolím.
Práce je metodickou přípravou na diplomovou práci. Součástí diplomové práce bude pomocí high-throughput sekvenačních metod analyzovat klinický materiál s cílem blíže popsat rozdíly v mykobiomu zdravých a nemocných jedinců. Vybrané druhy mykromycét průkazně asociované s mykobiomem nemocných jedinců budou kultivovány a charakterizovány na úrovni metabolizmu, produkce enzymů jakož i dalších fyziologických vlastností. Jejich vliv na imunitní odpověď bude studován in vitro na buněčných kulturách.

Preliminary scope of work in English

This Bachelor thesis should review background information about the mycobiome of human digestive tract and should serve as a basis of the Master thesis, which topic is specified below. This review should summarize the current knowledge about mycobiome of human intestine (incidence, interactions with other microbes, importance, ecology, relationship to the type of diet and various diseases). Intestinal mycobiota is considered an important factor in the development of certain diseases, which include inflammatory bowel disease (ulcerative colitis and Crohn's disease). The thesis will use high-throughput sequencing methods to analyze clinical material in order to further describe the differences in mycobiome of healthy and diseased individuals. Selected fungi significantly associated with diseased individuals will be cultured and characterized at the level of metabolism, production of enzymes and other physiological characteristics. Their influence on the immune response will be studied in vitro on cell cultures.
