Diplomová práce bude zaměřena na analýzu netěkavých N-nitrosaminů ve sladu. Tyto vysoce karcinogenní látky lze rozdělit do skupiny těkavých a netěkavých N-nitrosaminů. Těkavé N-nitrosaminy se běžně stanovují pomocí plynové chromatografie s hmotnostní či chemiluminiscenční detekcí. Netěkavé N-nitrosaminy dosud nelze spolehlivě a jednoduše stanovit, ale lze stanovit celkovou zdánlivou sumu všech N-nitroso sloučenin ve vzorku, tzv. ATNC. Cílem práce je vývoj nové metody stanovení netěkavých N-nitrosaminů v sladu, konkretně N-nitrosoprolinu a N-nitrososarkosinu.
Preliminary scope of work in English
This diploma thesis deals with analysis of N-nitrosamines in malt and beer. N-nitrosamines are compounds possessing a high carcinogenicity and can be classified as either volatile or non-volatile compounds. Volatile N-nitrosamines are routinely analyzed by gas chromatography coupled to mass spectrometry or chemiluminiscence detection techniques. Even these days, analysis of non-volatile N-nitrosamines is complicated and not always reliable. Usually the total amount of all N-nitroso compounds is determined in the sample and expressed as ATNC. The aim of this work is to develop a new and robust analytical method for analysis of non-volatile N-nitrosamines in malt, namely for N-nitrosoproline and N-nitrososarcosine.
