Studovali jsme využití testování cirkulujících nádorových buněk (CTC), tj. tekuté biopsie, u pacientů s kastračně rezistentním karcinomem prostaty (CRPC). Potřeba nového biomarkeru u těchto pacientů pramení především ze zavedení nových terapeutických možností, ale současnému nedostatku markerů umožňujícím lékařům jejich optimální výběr. Cílem studie bylo prozkoumat využití vzorků obohacených CTC získaných novou metodou AdnaTest (Quiagen, Německo), jako klinického biomarkeru u pacientů s CRPC. Chtěli jsme zavést metodu AdnaTest a vyhodnotit její charakteristiky na pacientských vzorcích. Plánovali jsme zjistit korelaci mezi detekcí CTC a klinicko-patologickými charakteristikami pacientů. Abychom prozkoumali možnosti analýzy CTC obohacených vzorků pomocí vysoko výkonné kvantitativné PCR a využití znalosti genové exprese v těchto vzorcích ke volbě terapie, chtěli jsme navrhnout a testovat nový multi-markerový panel pro monitorování charakteru CTC v průběhu terapie u pacientů trpících CRPC. |

We studied the implementation of circulating tumor cell (CTC) testing, i.e. liquid biopsy, in castration-resistant prostate cancer (CRPC) patients. The new biomarker is needed because of the currently enlarged portfolio of therapies and a lack of biomarkers available for therapeutical decision-making. The aim of this study was to explore the use of circulating tumor cell (CTC)-enriched samples obtained by the new method, AdnaTest (Qiagen, Germany), as a clinical biomarker in the castration-resistant prostate cancer (CRPC). We wanted to implement the AdnaTest method and to evaluate its characteristics on patient samples. We planned to correlate the CTC detection to the clinico-pathological characteristics of CRPC patients. In order to explore the use of CTC-enriched samples in high-throughput quantitative PCR analysis and to investigate the gene expression in CTC-enriched samples, we wanted to design and test a new multi-marker gene expression panel to monitor CTC character during CRPC therapy. |
