Denní rytmy jsou řízeny cirkadiánním pacemakerem umístěným v SCN. Astrocyty v SCN i jinde vykazují periodické denní změny v expresi genů a v akumulaci extracelulárního ATP. Předchozí studie prokázaly, že rytmus ATP je v obrácené fázi proti rytmu elektrické aktivity nebo proti rytmu AVP, což naznačuje, že ATP a AVP jsou uvolňovány z různých typů buněk a / nebo různými mechanismy. Cílem této diplomové práce je, zjistit vliv melatoninu na rytmus ATP u organotypických kultur suprachiasmatického jádra potkana a objasnit mechanismus tohoto případného vlivu.

Daily variation in physiology and behaviour are driven by circadian pacemakers; the SCN. Astrocytes in the SCN and elsewhere exhibit periodic daily fluctuations in gene expression and extracellular ATP accumulation. Previous studies have shown that rhythm ATP negatively correlates with electrical activity and arginine vasopressin (AVP) secretion rhythm, indicating that ATP and AVP are released from different cells and/or by different mechanisms. The aim of this study is to find out the effect of melatonin on ATP release from organotypic cultures on rat SCN and to clarify mechanism of this prospective effect.
