ATP syntáza je klíčovým enzymem celého energetického metabolismu. Nachází se ve vnitřní mitochondriální membráně, kde je součástí respiračního řetězce produkujícího energii ve formě adenosin trifosfátu. Regulace její aktivity je důležitá především kvůli udržování přiměřené hladiny ATP v buňce, která se mění dle aktuálních fyziologických nároků. Náhlé snížení aktivity ATP syntázy může vést k hyperpolarizaci mitochondriální membrány, spojené se zvýšenou produkcí reaktivních kyslíkových radikálů z respiračního řetězce. Takto navozený oxidativní stres může buňku fatálně poškodit a vést k programované buněčné smrti, apoptóze. Není proto překvapivé, že změny regulace aktivity ATP syntázy byly popsány u různých patologických stavů, jako například ischémie, nebo karcinogeneze. Cílem této práce je shrnout současné poznatky týkající se fyziologické regulace aktivity ATP syntázy, zejména na úrovni interakčních partnerů, Ca2+ iontů, post-translačních modifikací a v neposlední řadě i na úrovni supramolekulární organizace enzymu.

ATP synthase is a key enzyme of energy metabolism. It is located in the inner mitochondrial membrane, where it is a part of respiratory chain, which produces energy in the form of adenosine triphosphate. Regulation of its activity is important, primarily to maintain an adequate level of ATP in a cell, which is variable according to the current physiologicall demands. Sudden decrease in ATP synthase activity may lead to hyperpolarization of the mitochondrial membrane associated with increased production of reactive oxygen radicals from the respiratory chain. Oxidative stress induced in this way can fatally damage the cell and lead to a programmed cell death, apoptosis. It is therefore not surprising that changes in the regulation of ATP synthase activity have been described in various pathological conditions such as ischemia or carcinogenesis. The aim of this work is to summarize the current knowledge about the physiological regulation of ATP synthase activity, especially at the level of interaction partners, Ca2+ ions, post-translational modifications and, last but not least, at the level of supramolecular organization of the enzyme.
