Regulační paměťové T lymfocyty hrají zásadní roli v udržování autotolerance. Nicméně, interakce mezi regulačními T lymfocyty a autoreaktivními CD8+ T buňkami není zcele prostudována. Studentka použije myší model experimentálního autoimunitního diabetu indukovaného adoptivním transferem OVA-reaktivních T buněk do DEREG RIP-OVA myši (produkující OVA v pankreatických beta buňkách) a následnou imunizací OVA antigenem. DEREG RIP-OVA myši umožňují depleci regulačních T buněk po podání dipteriového toxinu. Porovnání myší, které mají Tregy a těch, ktereé je nemají, nám ukáže zda regulační T buňky zvyšují počet autoreaktivních T buněk, jejich affinitu k autoantigenu nebo množství autoantigenu, které jsou nutné pro indukci autoimunity.

Regulatory T cells play a crucial role in maintaining self-tolerance. However, the interaction between regulatory T cells and self-reactive CD8+ T cells are incompletely understood. The student will use our model of experimental autoimmune diabetes induced by the transfer of ovalbumin-reactive OT-I T cells into DEREG RIP-OVA mice (producing ovalbumin in pancreatic β cells) and subsequent priming of the OT-I T cells with their cognate antigen. The DEREG RIP-OVA mice enable depletion of regulatory T cells by the administration of diphtheria toxin. Comparison of T-reg-sufficient and Treg-deficient mice will show whether regulatory T cells increase number of self-reactive T cells, their affinity to the self-antigen, or the dose of the self-antigen required for the induction of autoimmunity.
