Protilátkami zprostředkovaná rejekce je těžká imunologická komplikace, která po transplantaci orgánů může ohrozit životy pacientů. Metodiky pro detekci a identifikaci HLA-specifických protilátek většinou nerozlišují, zda tyto protilátky vážou, či nevážou komplement. Cílem diplomové práce je proto zkoumat úlohu protilátek, které vážou komplement pro vznik imunologických komplikací u pacientů po transplantaci jater. Oddělení imunogenetiky IKEM má rozsáhlou databázi sér pacientů před a po transplantaci. Séra budou vyšetřena pomoci soupravy na přítomnost a specifikaci protilátek proti HLA antigenům I. a II třídy (Luminex) a potom za využití testu C1q (Onelambda) a C3d (Immucor) budou detekovány protilátky vážící komplement.

Antibody-mediated rejection is a severe immunological complication, which may endager patients lifes after organ transplantation. The methods for detection and identification of HLA-specific antibodies generally do not specify whether these antibodies are complement-binding or not. The aim of the diploma thesis is to study the role of antibodies which bind complement in relation to the development of immunological complications in patients after liver transplantation. The Department of immunogenetics IKEM has a large database of patient sera before and after transplantation. Sera will be tested by kits for detection and specification of antibodies to Class I and Class II HLA antigens (Luminex) and afterwards will be defined complement-binding antibodies using the C1q (OneLambda) and C3d (Immucor) kits.
